    Effects of propionate on streptolydigin production and carbon flux distribution in

    To achieve higher antibiotic streptolydigin productivity through metabolic regulation, propionate was fed during the fermentation of Streptomyces lydicus AS 4.2501. The effects of propionate feeding on streptolydigin production and intracellular fluxes were investigated. The highest streptolydigin production yield of 95.10mg•L-1 was obtained when 2mmol•L-1 of sodium propionate was added at 60h of cultivation into shake-flask culture. This yield is 23.06% higher when compared to that of a batch culture without propionate supplementation. It was also found that when propionate was added, much more organic acids were excreted. Metabolic flux analysis was performed and it demonstrated that the carbon fluxes of the pentose phosphate pathway and the anaplerotic reaction were significantly increased after propionate feeding. The carbon flux from pyruvate to acetyl-CoA was determined to be 24.7, which was 12.27% higher than that in the batch culture. This study indicated that the glucose-6-phosphate and pyruvate nodes were potential bottlenecks for increasing streptolydigin productivity. Potential targets and strategies that could be manipulated through genetic and process engineering to increase the production of streptolydigin were also suggested.

    A scheme of investigating the intracellular metabolic fluxes in central metabolism of Saccharomyces cerevisiae based on isotope model and tracer experiment was developed. The metabolic model applied in this study includes the Embden-Meyerhof-Parnas pathway, the pentose phosphate pathway, the tricarboxylic acid cycle, CO2 anaplerotic reactions, ethanol and acetate formation, and pathways involved in amino acid synthesis. The approach of hybridized genetic algorithm combined with the sequential simplex technique was used to optimize a quadratic error function without the requirement of the information on the partial derivatives. The impact of some key pa-rameters on the algorithm was studied. This approach was proved to be rapid and numerically stable in the analysis of the central metabolism of S.cerevisiae.

    A Lactobacillus brevis CGMCC 1306 isolated from fresh milk without pasteurization was found to have higher glutamate decarboxylase (GAD) activity. An effective isolation and purification procedure of GAD from a cell-free extract of Lactobacillus brevis was developed, and the procedure included four steps: 30%—90% saturation (NH4)2SO4 fractional precipitation, Q sepharose FF anion-exchange chromatography, sephacryl S-200 gel filtration, and resource Q anion-exchange chromatography. Using this protocol, the purified GAD was demonstrated to possess electrophoretic homogeneity via SDS-PAGE. The purification fold and activity recovery of GAD were 43.78 and 16.95%, respectively. The molecular weight of the purified GAD was estimated to be approximately 62 kDa via SDS-PAGE. The optimum pH and temperature of the purified GAD were 4.4 and 37℃, respectively. The purified GAD had a half-life of 50min at 45℃ and the Km value of the enzyme from Lineweaver-Burk plot was found to be 8.22. 5′-pyridoxal phosphate (PLP) had little effect on the regulation of its activity.

    The present study is focused on protein degradation during astaxanthin synthesis in Haematococcus pluvialis under high irradiance and nitrogen deficient conditions. It was found that with the onset of astaxanthin synthesis in the cultures of high light and nitrogen-free (HF), high light and nitrogen-repletion (HR), and low light and nitrogen-free (LF), (1) endopeptidase (EP) activities increased along with decrease in protein content, (2) asparagine in HF and HR rose significantly before the first 4 and 5 day, but fell after that time. While, it increased slowly and continuously in LF, (3) ammonium increased continuously in HF and HR, whereas in LF, it was detected on the sixth day, and increased slowly on the following days. By contrast, in low light and nitrogen-repletion culture, (LR), the contents of protein and asparagine as well as EP activity were maintained relatively constant, no astaxanthin and ammonium were detected. Furthermore, when HF was sealed and bubbled with CO2-free gas (O2 and N2), astaxanthin content increased as the protein level decreased. These results strongly suggest that (1) the degraded protein served as a substitutive carbon source, to some extent, for the biosynthesis of astaxanthin, (2) endopeptidase was involved in the degradative process, (3) for detoxification, part of the ammonium generated by protein degradation was transiently stored in asparagine, whereas the rest of it was expelled into the culture broth.
    Removal of di-n-butyl phthalate using immobilized microbial cells

    The biodegradation of di-n-butyl phthalate (DBP) using immobilized microbial cells was carried out in an internal airlift loop reactor with ceramic honeycomb supports. A strain that is capable of degrading DBP was isolated from the activated sludge and identified as Bacillus sp. using 16S rDNA sequential analysis. Bacillus sp. could be rapidly attached onto the ceramic honeycomb supports. The immobilized cells could effectively degrade DBP in batch and continuous experiments. When the influent concentration of DBP was 50mg•L-1, the effluent DBP reached less than 1mg•L-1 with 6h hydraulic retention time (HRT) in continuous experiment. The immobilized microbial cells could grow and accumulate through the biodegradation of DBP, and the rate of degradation is accordingly increased. The possible pathway of DBP biodegradation using immobilized cells was tentatively proposed.

    A genetically engineered Escherichia coli JM109 harboring pLF3 was used to produce a hybrid extracellular -glucanase. Starting with enzyme production medium, glycerol and yeast extract combined with NaNO3 were screened to be the most suitable carbon and nitrogen source, respectively. Analysis of six components of the enzyme production medium by employing statistical optimization methods such as Plackett-Burman design and steepest ascent showed that yeast extract was the only significant variable and its best concentration for enzyme production was 12g•L-1. After optimization of the medium, 297.71U•ml-1 of β-glucanase activity in the medium and 352350U•g-1 of β-glucanase selectivity could be obtained, which were 14 and 72 folds higher than those ob-tained from original medium, respectively. Even higher enzyme activities were achieved by batch cultivations in a conventional stirred bioreactor on the optimized medium.

    Titania catalysts were synthesized by a solution combustion method (SCM). Photodegradation of 4-chlorophenol (4-CP) using the synthesized catalysts was studied under both visible light (λ≥420nm) and sunlight irradiation. The effect of preparation conditions on photocatalytic activities of the synthesized catalysts was investigated. The optimal photocatalytic activity of the catalyst(denoted as A1)was obtained under the following synthesis conditions: ignition temperature of 350℃, fuel ratio ( ) of 1 and calcination time of 1h. The degradation and mineralization ratio of 4-CP were 78.2% and 53.7% respectively under visible light irradiation for 3h using catalyst A1. And the catalyst A1 also showed high photocatalytic activity under sunlight irradiation.

    The thermal decomposition of potassium titanium hexacyanoferrate(Ⅱ) (KTiFC) loaded with cesium (referred to as Used Exchanger, or UE) was studied at different flow rate of air in a fixed bed calciner. The calcination process consisted of four stages: ambient temperature—180℃ (stage Ⅰ), 180—250℃ (stage Ⅱ), 250—400℃ (stage Ⅲ), and constant 400℃ (stage Ⅳ). The most intense reaction occurred in stage Ⅱ. The rate of thermal decomposition was controlled, depending on the O2 flux, by O2 or CN concentration in different stages. Results from differential thermal analysis (DTA) showed that the calcination reaction of the anhydrous UE was exothermic, with an approximate heat output of 4.6kJ•g-1, which was so large to cause the possible agglomeration of calcined residues. The agglomeration could be avoided by enhancing heat transfer and controlling the O2 flux. It was found that there was no cyanides in the calcined residues and no CN-bearing gases such as HCN and (CN)2 in the off-gas. It seemed that the catalytic oxidation furnace behind the fixed bed calciner could be cancelled.

    A magnetically separable photocatalyst TiO2/SiO2/NiFe2O4 (TSN) with a typical ferromagnetic hysteresis was prepared by a liquid catalytic phase transfer method. When the intensity of applied magnetic field weakened to zero, the remnant magnetism of the prepared photocatalyst faded to zero. The photocatalytst can be separated from water when an external magnetic field is added and  redispersed into aqueous solution after the external magnetic field is eliminated, that makes the photocatalysts promising for wastewater treatment. Transmission electron microscope (TEM) and X-ray diffractometer (XRD) were used to characterize the structure of the photocatalyst, indicating that the magnetic SiO2/NiFe2O4 (SN) particle was compactly enveloped by P-25 titania and TiO2 shell was formed. The magnetic composite showed high photocatalytic activity for the degradation of methyl orange in water. A thin SiO2 layer between NiFe2O4 and TiO2 shell prevented effectively the leakage of charges from TiO2 particles to NiFe2O4, which gave rise to the increase in photocatalytic activity. Moreover, the experiment on recycled use of TSN demonstrated a good repeatability of the photocatalytic activity.

    Epoxidation of cyclohexene to cyclohexene oxide was studied in a new type reactor—the ultrasound airlift loop reactor. The influences of ultrasound intensity, molar ratio of isobutyraldehyde to cyclohexene and oxygen gas flow rate on the conversion of cyclohexene and selectivity of cyclohexene oxide were investigated and discussed, and the optimal operation condition was found, under which 95.2% conversion of cyclohexene and 90.7% selectivity of cyclohexene oxide were achieved. The ultrasonic airlift loop reactor utilizes the synergistic effect of sonochemsitry and higher oxygen transfer rate. Possible reaction mechanisms were outlined and the reason of ultrasound promotion of epoxidation reactionwas analyzed.

    The aim of this work is to investigate the flow instabilities in a baffled, stirred tank generated by a single Rushton turbine by means of large eddy simulation (LES). The sliding mesh method was used for the coupling between the rotating and the stationary frame of references. The calculations were carried out on the “Shengcao-21C” supercomputer using a computational fluid dynamics (CFD) code CFX5. The flow fields predicted by the LES simulation and the simulation using standard k-ε model were compared to the results from particle image ve-locimetry (PIV) measurements. It is shown that the CFD simulations using the LES approach and the standard k-ε model agree well with the PIV measurements. Fluctuations of the radial and axial velocity are predicted at different frequencies by the LES simulation. Velocity fluctuations of high frequencies are seen in the impeller region, while low frequencies velocity fluctuations are observed in the bulk flow. A low frequency velocity fluctuation with a nondimensional frequency of 0.027Hz is predicted by the LES simulation, which agrees with experimental investi-gations in the literature. Flow circulation patterns predicted by the LES simulation are asymmetric, stochastic and complex, spanning a large portion of the tanks and varying with time, while circulation patterns calculated by the simulation using the standard k-ε model are symmetric. The results of the present work give better understanding to the flow instabilities in the mechanically agitated tank. However, further analysis of the LES calculated velocity se-ries by means of fast Fourier transform (FFT) and/or spectra analysis are recommended in future work in order to gain more knowledge of the complicated flow phenomena.

    In this study, extraction of p-cresol from the simulated wastewater has been studied using centrifugal extractors and QH-1 extractant. The distribution ratio of p-cresol was approximately 308.5 in the QH-1-p-cresol (pH=2.0) system. The extraction stage efficiency of the single-stage centrifugal extractor was higher than 92%, and the extraction rate of the three-stage cascade was higher than 99%. When 15% NaOH was used for the stripping of p-cresol in QH-1 solution, the stripping rate of the three-stage cascade reached 100%. A mathematical model of multistage countercurrent extraction process that considers the phase volume change, extraction stage efficiency, and phase entrainment was used to simulate the extraction of p-cresol using centrifugal extractors. The calculated results using this model were in good agreement with the experimental results.

    Measurement and correlation for solubility of dimethyl-2,6-naphthalene dicarboxylate in

    Solubility of dimethyl-2,6-naphthalene dicarboxylate in acetic acid, N,N-dimethylformamide, N,N-dimethyl acetamide, dimethyl sulphoxide, and N-methyl-2-ketopyrrolidine were determined using a dynamic method. The measured systems were correlated by UNIFAC group contribution method. A new main group (aromatic ester, ACCOO) was defined to express the activity coefficients of the aromatic ester. New interaction parameters of the ACCOO group were expressed as the first-order function of temperature and were determined from the experimental data. The calculated results for the new interaction parameters were satisfactory. The measured systems were also correlated with the Wilson and λ-h models, and the results were compared with those of the UNIFAC model.
    Modeling VLE and GLE of systems involving polymers by using SRK equation of state

    A simple extension of cubic equations of state (EOS) to polymer systems has been proposed. The So-ave-Redlich-Kwong (SRK) EOS was taken as a prototype to be used to describe the PVT behavior of polymer melts in a wide temperature and pressure range. Combined with a modified Huron-Vidal gE-mixing rule it was applied for modeling vapor-liquid equilibria of polymer-solvent solutions and the solubility of supercritical gases in polymer melts. Satisfactory results are obtained.

    By using the laser-monitoring technique, solid-liquid equilibrium data of glutaric acid in cyclohexanol, cyclohexanone, their five mixed solvents and acetic acid were measured within the temperature range from 292.15K to 354.60K by dynamic method. Empirical formula and λh equation were used to correlate the solubilities of glutaric acid in eight solvents. The maximal average relative deviations were 1.15% and 2.20% by using the em-pirical formula and λh equation to correlate the solubility data. The results showed that the empirical formula and λh model could correlate the solubility data of glutaric acid in eight solvents. In addition, the solubility data of glutaric acid in five mixtures (cyclohexanone+cyclohexanol) could be predicted with the NRTL equation utilizing the pa-rameters of the binary systems. The total average relative deviation was 3.60%. The results indicate that the NRTL equation could well predict the solubilities of glutaric acid in the mixed solvents of cyclohexanone and cyclohexanol.

    This study describes a classification methodology based on support vector machines (SVMs), which offer superior classification performance for fault diagnosis in chemical process engineering. The method incorporates an efficient parameter tuning procedure (based on minimization of radius/margin bound for SVM’s leave-one-out errors) into a multi-class classification strategy using a fuzzy decision factor, which is named fuzzy support vector machine (FSVM). The datasets generated from the Tennessee Eastman process (TEP) simulator were used to evaluate the classification performance. To decrease the negative influence of the auto-correlated and irrelevant variables, a key variable identification procedure using recursive feature elimination, based on the SVM is implemented, with time lags incorporated, before every classifier is trained, and the number of relatively important variables to every classifier is basically determined by 10-fold cross-validation. Performance comparisons are implemented among several kinds of multi-class decision machines, by which the effectiveness of the proposed approach is proved.

    A splicing system based genetic algorithm is proposed to optimize dynamical radial basis function (RBF) neural network, which is used to extract valuable process information from input output data. The novel RBF network training technique includes the network structure into the set of function centers by compromising between the conflicting requirements of reducing prediction error and simultaneously decreasing model complexity. The ef-fectiveness of the proposed method is illustrated through the development of dynamic models as a benchmark discrete example and a continuous stirred tank reactor by comparing with several different RBF network training methods.

    By extending the system’s state variables, a novel predictive functional controller has been  developed. The structure of this controller is similar to that of classical proportional integral (PI) optimal controller and includes a control block that can perform a feed-forward control of future P-step set points. It considers both the state variables and the output errors in its cost function, which results in enhanced control performance compared with traditional state space predictive functional control (TSSPFC) methods that consider only the predictive output er-rors. The predictive functional controller (PFC) has been compared with TSSPFC in terms of tracking ability, dis-turbance rejection, and also based on its application to heavy oil coking equipment. The results obtained show the effectiveness of the controller.
    A simultaneous optimization method for the structure and composition differences of a mass  exchange network

    This article presents a new method to calculate the composition differences (ε) for targeting the minimum total annualized cost (TAC) of a mass exchange network (MEN), which is based on the combination of composition interval diagram (CID) with mathematical programming. The total cost target consists of the capital cost of the process units and the operating cost for mass separating agents (MSAs). The value of total cost varies considerably with the composition differences, so the values of ε should be optimized in order to obtain minimum TAC of a MEN. This article considers ε as a set of unequal variables for each equilibrium equation of a rich-lean stream pair, employing them to build the CID and mathematical model, which optimizes the structure and composition differ-ences simultaneously. Two examples are applied to illustrate the proposed method and the results demonstrate that the approach introduced by this article is simpler and more convenient than the methods in previous literatures.

    NiOOH was prepared by one-step electrolysis of spherical Ni(OH)2 and the effects of electrolysis parameters were examined. The highly pure NiOOH was obtained after electrolysis at a current density of 60mA•g-1 and 30℃ with anodic potential controlled in the range of 1.73—1.85V (vs. Zn/ZnO) for 360min. The NiOOH samples were characterized by X-ray powder diffraction (XRD) and scanning electron microscope (SEM) analysis. Results indicate that the electrolysis product is spherical NiOOH doped with graphite. Charge and discharge tests show that the prepared NiOOH offers a discharge capacity of over 270mAh•g-1 at current density of 30mA•g-1 and can be directly used as cathode material of alkaline Zn/NiOOH batteries. Galvanostatic charge/discharge and cyclic volt-ammetry (CV) tests reveal good cycling reversibility of the NiOOH electrode.
    Species transformation of trace elements and their distribution prediction in dyestuff

    The release of heavy metals from the combustion of hazardous wastes is an environmental issue of increasing concern. The species transformation characteristics of toxic heavy metals and their distribution are considered to be a complex problem of mechanism. The behavior of hazardous dyestuff residue is investigated in a tubular furnace under the general condition of hazardous waste pyrolysis and gasfication. Data interpretation has been aided by parallel theoretical study based on a thermodynamic equilibrium model based on the principle of Gibbs free energy minimization. The results show that Ni, Zn, Mn, and Cr are more enriched in dyestuff residue incineration than other heavy metals (Hg, As, and Se) subjected to volatilization. The thermodynamic model calculation is used for explaining the experiment data at 800℃ and analyzing species transformation of heavy metals. These results of species transformation are used to predict the distribution and emission characteristics of trace elements. Although most trace element predictions are validated by the measurements, cautions are in order due to the complexity of incineration systems.

    Alkalinity is one of the most important parameters that influence microbial metabolism and activity during sulfate-laden wastewater biological treatment. To comprehensively understand the structure and dynamics of functional microbial community under alkalinity changes in sulfate-reducing continuous stirred tank reactor (CSTR), fluorescent in situ hybridization (FISH) technique was selected for qualitative and semi-quantitative analysis of functional microbial compositions in activated sludge. During 93d of bioreactor operation, the influent alkalinity was adjusted by adding sodium bicarbonate from 4000mg•L-1 down to 3000mg•L-1, then to 1500mg•L-1, whereas other parameters, such as the loading rates of chemical oxygen demand (COD) and sulfate ( ), hy-draulic retention time (HRT), and pH value, were continuously maintained at 24g•L-1•d-1 and 4.8g•L-1•d-1, 10h, and about 6.7, respectively. Sludge samples were collected during different alkalinity levels, and total Bacteria, the sulfate-reducing bacteria (SRB), and four SRB genera were demonstrated with 16S ribosomal RNA-targeted oli-gonucleotide probes. The results indicated that bioreactor started-up successfully in 30d. The two instances of drop in alkalinity resulted in the fluctuation of sulfate removal rate. The diversity of SRB community showed significant shift, and the alteration of microbial community directly resulted in the corresponding statuses of bioreactor. The dominant genera during the bioreactor start-up and alkalinity drops were Desulfovibrio, Desulfobacter, Desulfovi-brio, Desulfobacter, and Desulfovibrio, respectively. In addition, the acetotrophic SRB suffered more from the re-duction of alkalinity than the non-acetotrophic SRB. This strategy can present the functional microbial community structure during start-up and alkalinity drop stages, and provides a powerful theoretical guideline for optimization and adjustment of bioreactor, as well.
    Wet gas meter development based on slotted orifice couple and neural network techniques

    A slotted orifice has many superiorities over a standard orifice. For single-phase flow measurement, its flow coefficient is insensitive to the upstream velocity profile. For two phase flow measurement, various characteristics of its differential pressure (DP) are stable and closely correlated with the mass flow rate of gas and liquid. The complex relationships between the signal features and the two-phase flow rate are established through the use of a back propagation (BP) neural network. Experiments were carried out in the horizontal tubes with 50mm inner di-ameter, operated with water flow rate in the range of 0.2m3•h-1 to 4m3•h-1, gas flow rate in the range of 100m3•h-1 to 1000m3•h-1, and pressure at 400kPa and 850kPa respectively, where the temperature is ambient temperature. This article includes the principle of wet gas meter development, the experimental matrix, the signal processing techniques and the achieved results. On the basis of the results it is suggested that the slotted orifice couple with a trained neural network may provide a simple but efficient solution to the wet gas meter development.
    Design and operation of batch extractive distillation with two reboilers

    This paper proposes a modified operation mode of batch extractive distillation with two reboilers. The separation of ethanol and water using as ethylene glycol is well done to validate the feasibility of this mode. The pilot-plant experimental data under the identical operational conditions between the regular mode and the modified mode are in particular discussed. The studied results prove more practical advantages of the modified mode over the regular mode such as easier operation control, more flexibility and higher separation efficiency etc. So it supplies certain guidance for the industrial application of batch extractive distillation process.

    In reversed-phase high performance liquid chromatography (RP-HPLC), the mobile phase condition for separating eight isoflavones (daidzin, glycitin, genistin, 6′-o-acetyl daidzin, 6′-o-malonyl genistin, daidzein, glycitein and genistein) was optimized using the HCI (High-Purity Separation Laboratory, Department of Chemical Engineering, Inha University) program software. The optimum composition of mobile phase for the separation of the eight isoflavones was obtained. The elution profiles were calculated by the plate theory based on the equations of retention factor, ln k=A+BF+CF2, where F was the volume percentage of acetonitrile with 0.1% acetic acid (AA). The first mobile phase composition was water with 0.1% AA/acetonitrile with 0.1% AA (88%/12%, by volume), followed at 9min later by the second composition of mobile phase which was step-changed to 85%/15%, at 19min by the third composition which was step-changed to 73%/27%, at 30min when it was changed to 65%/35% and finally it was maintained in isocratic mode to the end of the run time at 50min. Although, using step gradient mode to separate the isoflavones, the calculated and experimented data were not achieved very good agreement, we could estimate the closed retention time before experiment. And the agreement between the experimental data and the calculated values was relatively good using isocratic separation for eight isoflavones, but the retention time is very long.

    For the optimal design of a heat exchanger network, the inlet and outlet stream temperatures of each heat exchanger in the network should be known. An explicit analytical solution of stream temperatures of an arbitrary connected heat exchanger network was introduced, which is suitable for the thermal calculation of heat exchanger networks. For the heat exchanger network synthesis, this solution was further developed and coupled with the stage-wise superstructure heat exchanger networks. The new calculation procedure reduced the computer memory requirement dramatically. On the basis of this solution, a mathematical model for synthesis of heat exchanger networks with genetic algorithm was formulated, which is always feasible and no iteration is needed. Two examples were calculated with the proposed approach and better results were obtained.

    A novel thermosetting resin system for superabrasives based on novolak and bismaleimide (BMI) was developed. The novolak resin was allylated and then copolymerized with BMI. The structure of allyl novolak and reaction mechanism were analyzed by FTIR. Thermal and mechanical properties were characterized by using com-prehensive thermal analyzer (DSC-TG) and strength tester, respectively. The results showed that high molecular weight of novolak was advantageous for heat-resistance, but was unfavorable for the bending strength. High allyl content improved the heat-resistance but lowered the bending strength. When the molecular weight of novolak was 450 and allyl content was 50%, the best resin system with good heat-resistance and bending strength was obtained. It was suitable for the manufacturing of superabrasive tools.
